The Galleria Borghese is a must-see sight that I learned about from Company of Men.  Reservations are required. 

I did a 1-day private tour on a golf cart around Rome last year, and it was definitely money well spent for a group of 3 of us.  It included front of the line access to the Vatican, so we got to see the Sistine Chapel and St Peter's Basilica.

The Castle Santa Angelo was a nice day (lots of stair climbing).

The Museum Capiolini was a nice day with lots of statues and artwork.

I didn't tour the inside of the Colosseum because of the crowds.  But I don't feel I missed much considering all the other sites I saw on my short trip last year.

My favorite dish in Rome was Pasta Carbonara.  It's completely different than the the creamy dish served in America.  I tried it at about 5 different restaurants, and it was always great.
