Charleston is a lovely old town ..but sadly in the middle of nowhere.

Everywhere is in the middle of nowhere to some extent. Barry Humphries (aka Dame Edna) once remarked that Australia is soooo central, why you can hop on a plane and in just 24 hours be somewhere interesting (NYC is 20 hours away). Arguably, if you subscribe to the NYC hype (not completely unwarranted) it is in the middle of nowhere because the rest of the world is a void by comparison. I'm often content to enjoy the place that I'm in for itself, not for what it's in the middle of. Charleston is on my list, that it is in South Carolina doesn't change that.

Actually...the comparison between NYC and Charleston is vast. My wife and I drive out of town most every weekend and there are hundreds of interesting things to do just outside of NYC.  There are multiple significant municipalities just beyond the city limits like Scarsdale and Huntington that are beautiful charming villages.

Sadly Charleston for all it's beauty sits there alone as a destination. As far as being gay and living there... I imagine you'd be very happy in the company of your lover but somewhat lonely if you try to live the typical bar and club social life of the single urban gay man.

I've considered retiring there myself but the lack of any culture surrounding the city always makes Charleston an impractical alternative.

between Orlando and Charlotte (which I realize is not Charleston but up the road a piece).

That's a damn big "piece".  Charleston and Charlotte have to be at least 200 miles from each other.

Seriously doubt you're gonna get an escort to drive four hours each way for a hire, unless you're VERY generous.
